摘要
采用6-氨基喹啉-N-(羥基琥珀酰亞胺基)氨基甲酸酯(AQC)為柱前衍生化試劑,建立了AQC柱前衍生化反相高效液相色譜法用于測定大蒜中的蒜氨酸及其有關物質(zhì)的含量。

該衍生化反應可瞬間完成,衍生化產(chǎn)物穩(wěn)定。采用的色譜條件為:KromasilC18柱(250mm×4.6mm,5μm),流動相A為0.1%乙酸銨(含0.03%乙酸),流動相B為水-乙腈(體積比為40∶60),線性梯度洗脫,流速為1.0mL/min,檢測波長為248nm。蒜氨酸在1.1719~1500μg/mL范圍內(nèi)線性關系良好(r=0.9998),日內(nèi)、日間測定的精密度良好(相對標準偏差(RSD)為1.8%,n=5),添加回收率為99.1%(RSD=1.9%,n=5),最小檢出濃度為0.15μg/mL。該方法準確、方便、快速。
