反相高效液相色譜法(RP-HPLC)測定異構顆粒酒花中的異α-酸
內容
利用反相高效液相色譜法(RP-HPLC)研究異構顆粒酒花中異α-酸的測定方法。結果表明:當檢測波長270nm、流動相為甲醇:水:磷酸(85%):EDTA(ethylene diamine tetra aceticacid)二鈉鹽(0.2mol/L)(770:210:5:1)、流速為1.0ml/min時,方法的精密度為1.09%,外加標回收率91.4%~97.1%之間。RP-HPLC的色譜柱為EC250/4Nuclesoil,100-5C18(4.0×250nm,5μm),保護柱為CC8/4Nuclesoil100-5C18。該方法可以簡便、準確地測定異構顆粒酒花中異α-酸含量。
Iso-α-acids content of isomerized hop pellets was analyzed by reversed-phase high performance liquid chroma- tography (RP-HPLC). The optimized conditions are: 270 nm by ultraviolet detection with methanol: H2O: H3PO4 (85%): EDTA- Na2 (0.2 mol/L) (770: 210: 5: 1, respectively) as mobile phase with flow rate 1.0 ml/min.
