冷卻豬肉用1 mol.L-1高氯酸水溶液勻漿去除蛋白質(zhì)及脂肪后,5000g離心,并過(guò)0.25μm纖維素膜,然后以高效液相色譜反相柱分離,通過(guò)紫外檢測(cè)器檢測(cè)。檢測(cè)結(jié)果顯示,該測(cè)定方法回收率為95.5%~102.3%,相對(duì)標(biāo)準(zhǔn)偏差為3.2%~5.7%,日內(nèi)測(cè)定變異系數(shù)(CV)為1.09%,日間測(cè)定CV為1.28%。該方法應(yīng)用于冷卻豬肉乳酸含量測(cè)定獲得較好的效果。
A reversed-phase high performance liquid chromatography(RP-HPLC)method was established to determine lactic acid content in chilled meat.The samples pretreated with the solution of 1 mol·L-1 perchloric acid(PCA)was separated through RP-HPLC.The determined result indicated that the recovery rate was 95.5%-102.3%,and relative standard deviation was 3.2%-5.7%,The coefficient of variance within-day and between-day was 1.09% and 1.28% respectively.The method was successfully used in the analysis of chilled pork.
