目的:建立HPLC法測(cè)定果汁中9種有機(jī)酸的檢測(cè)方法。

果汁中9種有機(jī)酸的反相高效液相色譜法測(cè)定
方法:果汁樣品經(jīng)稀釋過濾后,采用Gemini C18色譜柱(4.6 mm×250 mm,5μm)分離,以0.01 mol/L磷酸氫二銨溶液(用磷酸調(diào)節(jié)pH=2.6)作為流動(dòng)相,UV210 nm檢測(cè),外標(biāo)法定量。結(jié)果:9種有機(jī)酸的線性范圍分別為草酸1.0~400 mg/L、酒石酸2.0~500 mg/L、丙酮酸1.0~500 mg/L、蘋果酸5.0~2000 mg/L、抗壞血酸1.0~400 mg/L、乙酸8.0~2000 mg/L、檸檬酸5.0~2000 mg/L、富馬酸0.5~100 mg/L、反烏頭酸1.0~400 mg/L,各種有機(jī)酸的線性相關(guān)系數(shù)r>0.999,回收率在93.7%~109.0%,RSD均小于5.0%,最低檢測(cè)限在0.2~7.5 mg/L范圍。結(jié)論:該方法具有快速、靈敏、簡(jiǎn)便、準(zhǔn)確等優(yōu)點(diǎn),可應(yīng)用于果汁中有機(jī)酸的檢測(cè),能滿足實(shí)際工作的需要。
